TBXT (Brachyury) is a T-box family DNA-binding transcription factor with a central role in mesoderm specification and notochord formation during embryonic development. Through its conserved T-box DNA-binding domain, Brachyury binds specific T-site sequences and regulates genes required for mesoderm formation and differentiation. Its expression is closely associated with notochord-derived cells and it is predominantly localised to the nucleus.
TBXT is also an important research target in chordoma, a tumour arising from notochordal remnants, where Brachyury expression is strongly associated with notochordal differentiation and has been investigated as a molecular driver of tumour biology. Beyond chordoma, aberrant Brachyury expression has been studied in epithelial–mesenchymal transition, tumour-cell plasticity, invasion and treatment-response mechanisms across a range of cancer models.
This makes TBXT/Brachyury relevant to research in embryonic development, mesoderm and notochord biology, transcriptional regulation, chordoma biology and cancer cell-state transitions.
