Built around the experiment

Your project. A defined technical scope.

Antibody engineering adapts an existing clone or sequence to a new experimental requirement. ABMIUM coordinates projects covering hybridoma sequencing, recombinant expression, antibody format changes and humanisation. The starting sequence, binding assay and required final format determine the scope. Agree how binding and function will be compared with the original material before choosing an engineering strategy.

Specialist ProMab service routes can be coordinated through ABMIUM. Your quotation identifies the selected provider, agreed stages, available documentation and material or experimental deliverables.

ABMIUM reviews your specification and coordinates suitable custom-service and manufacturing routes. The proposed provider or manufacturer, available documentation and agreed scope are stated before order.

Technical scope

Request identification of antibody variable-region sequences from a supplied hybridoma. Confirm sample condition, clonality, the number of chains to resolve and the desired sequence deliverable. Sequence recovery can support recombinant conversion, construct development and continuity of a research antibody.

Technical scope

Convert a characterised antibody sequence into an agreed recombinant format such as IgG, Fab, scFv or an Fc fusion. Define species, isotype, tags and purification requirements. Expression and binding checks should be scoped together so the new format can be compared with the original reagent.

Technical scope

Discuss humanisation of non-human variable regions with a plan to assess retention of binding and activity. Provide the parental sequence and any existing functional data. A humanised sequence is an engineered research candidate; clinical suitability is a separate development question.

Technical scope

Explore affinity optimisation against a defined antigen and assay objective. State whether the goal is improved binding, altered kinetics or preserved specificity. Research-scale expression, purification and material supply can be added, with QC and comparison assays defined in the quotation.

Clear answers before you enquire

Frequently asked questions

Can you sequence an existing hybridoma?

Hybridoma sequencing can be scoped through ABMIUM. Provide cell-line details and sample availability so the route and chain-resolution requirements can be assessed.

Can an IgG be converted to an scFv?

Reformatting can be discussed from an antibody sequence. Binding may change with format, so expression and functional comparison should be part of the project plan.

Does humanisation guarantee unchanged binding?

No. Retention of binding and function needs evaluation against the parental antibody. The quotation should state the design and validation stages.

Can engineering and production be quoted together?

Yes. Include the final amount, buffer, purity, application and QC requirements so the engineering and material-supply stages can be scoped together.

How does antibody humanisation (humanization) differ from recombinant conversion?

No. Recombinant conversion expresses a sequence-defined antibody; humanisation changes sequence composition toward a human framework while seeking to retain useful binding. Provide the parent sequence or source material, intended format and functional assay so the engineering and comparison studies can be scoped separately.

What should be tested after antibody engineering?

Compare the engineered material with the parent using binding and relevant functional assays. Expression, aggregation and stability may also influence whether a variant is useful. Define the comparator, acceptance criteria and requested analysis before selecting an engineering package.

What affects the cost of antibody engineering & humanisation?

Pricing depends on starting sequence or source material, number of variants, expression format and binding or functional comparisons. ABMIUM provides a project-specific quotation after reviewing the requirement. Ask for a breakdown of stages, deliverables, optional studies and further-work decision points so you can compare the total scope rather than an entry price alone.
