Product description

Background

Nuclear hormone receptor. Binds estrogens with an affinity similar to that of ESR1, and activates expression of reporter genes containing estrogen response elements (ERE) in an estrogen-dependent manner (PubMed: 20074560). Isoform beta-cx lacks ligand binding ability and has no or only very low ere binding activity resulting in the loss of ligand-dependent transactivation ability. DNA-binding by ESR1 and ESR2 is rapidly lost at 37 degrees Celsius in the absence of ligand while in the presence of 17 beta-estradiol and 4-hydroxy-tamoxifen loss in DNA-binding at elevated temperature is more gradual.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human ESR2 antibody. ESR2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human ESR2 Antibody is added and binds to ESR2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ESR2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human ESR2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.
