Product description

Background

Substrate recognition component of a SCF-like ECS (Elongin BC-CUL2/5-SOCS-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins (PubMed: 15601820, PubMed: 21199876). Negatively regulates nitric oxide (NO) production and limits cellular toxicity in activated macrophages by mediating the ubiquitination and proteasomal degradation of NOS2 (PubMed: 21199876). Acts as a bridge which links NOS2 with the ECS E3 ubiquitin ligase complex components ELOC and CUL5 (PubMed: 21199876).

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human SPSB1 antibody. SPSB1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human SPSB1 Antibody is added and binds to SPSB1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SPSB1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human SPSB1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.
