Product description

Background

Growth factor of the TGF-beta superfamily that plays essential roles in many developmental processes including cartilage and bone formation (By similarity). Plays also an important role in the regulation of iron metabolism by acting as a ligand for hemojuvelin/HJV (PubMed:19252486). Initiates the canonical BMP signaling cascade by associating with type I receptor ACVR1 and type II receptor ACVR2B. In turn, ACVR1 propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target. Can also signal through non-canonical pathway such as TAZ-Hippo signaling cascade to modulate VEGF signaling by regulating VEGFR2 expression (By similarity).

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse BMP6 antibody. BMP6 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse BMP6 Antibody is added and binds to BMP6 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BMP6 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse BMP6. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

BMP6bone morphogenetic protein 6

View target guide

Bone morphogenetic protein 6 (BMP6/BMP-6) is a secreted TGF-beta-superfamily growth factor involved in developmental, skeletal and iron-homeostasis signalling. ABMIUM catalogue products linked below support research on BMP6 biology.

Biological function: BMP6 is a TGF-beta-superfamily ligand that promotes cartilage and bone formation and initiates canonical BMP signalling through receptor complexes including ACVR1 and ACVR2B, activating SMAD1/5/8. Reviewed UniProt also establishes an important role in HAMP/hepcidin regulation and systemic iron metabolism, including signalling linked to hemojuvelin (HJV).
