AKT1S1 (AKT1 substrate 1; PRAS40) encodes proline-rich AKT1 substrate 1, a phosphorylation-sensitive regulator of mechanistic target of rapamycin complex 1 (mTORC1).
Biological function: PRAS40/AKT1S1 negatively regulates mTORC1 by associating with the complex and limiting substrate access under low insulin or nutrient conditions. Insulin- and nutrient-responsive phosphorylation promotes 14-3-3 binding and relief of mTORC1 inhibition.
