Test principle

HBcAg is used to coat the reaction plate. The test specimen is then added, along with HBcAb-HRP. When HBcAb is present in the specimen, it competes with HBcAb-HRP for binding to the HBcAg coated on the plate, forming an HBcAg-HBcAb-HRP complex. The addition of TMB substrate triggers acolorimetric reaction; the lower the concentration of HBcAb, the higher the OD value; conversely, the higher the concentration of HBcAb, the lower the OD value. The presence of HBc –Ab can be determined by measuring the absorbance value by using a microplate reader with 450 nm (630 nm) wavelength.
