Le kit ELISA humain CA12 (Anhydrase Carbonique XII) est un kit ELISA d'ELK Biotechnology pour la mesure de recherche de CA12.
Le principe du test appliqué dans ce kit est l'immunodosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-revêtue d'un anticorps spécifique de la CA12 humaine. Des étalons ou des échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps biotine-conjugué spécifique de la CA12 humaine. Ensuite, de l'avidine conjuguée à la peroxydase de raifort (HRP) est ajoutée à chaque puits de la microplaque et incubée. Après l'ajout de la solution de substrat TMB, seuls les puits contenant de la CA12 humaine, l'anticorps biotine-conjugué et l'avidine conjuguée à l'enzyme présenteront un changement de couleur. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré par spectrophotométrie à une longueur d'onde de 450 nm ± 10 nm. La concentration de CA12 humaine dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.

Carbonic anhydrase 12 (CA12; carbonic anhydrase XII/CA-XII) is a type I transmembrane zinc metalloenzyme with an extracellular catalytic domain. It is studied in epithelial CO2/bicarbonate handling, extracellular pH regulation and tissue-specific membrane carbonic-anhydrase biology.
Biological function: CA12 catalyses reversible hydration of carbon dioxide at the cell surface, generating bicarbonate and protons and contributing to extracellular acid-base handling. Its transmembrane localisation links carbonic-anhydrase catalysis to epithelial transport and pH-regulatory systems.
