Product description

Background

Aldehyde dehydrogenase, mitochondrial is an enzyme that in humans is encoded by the ALDH2 gene located on chromosome 12. This protein belongs to the aldehyde dehydrogenase family of enzymes. Aldehyde dehydrogenase is the second enzyme of the major oxidative pathway of alcohol metabolism. Two major liver isoforms of aldehyde dehydrogenase, cytosolic and mitochondrial, can be distinguished by their electrophoretic mobilities, kinetic properties, and subcellular localizations.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse ALDH2 antibody. ALDH2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse ALDH2 Antibody is added and binds to ALDH2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ALDH2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse ALDH2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

ALDH2aldehyde dehydrogenase 2 family member

View target guide

ALDH2 (aldehyde dehydrogenase 2 family member), also known as mitochondrial aldehyde dehydrogenase, is a mitochondrial matrix enzyme that oxidises acetaldehyde and other aldehydes. Researchers use ALDH2 to study aldehyde metabolism, alcohol metabolism and mitochondrial responses to carbonyl stress.

Biological function: ALDH2 is the mitochondrial aldehyde dehydrogenase isoform with low affinity constant for acetaldehyde and is localised to the mitochondrial matrix. It catalyses a major step in oxidative alcohol metabolism by converting acetaldehyde to acetate.
