Product description

Background

[Isoform 1]: A non-specific tyrosine phosphatase that dephosphorylates a diverse number of substrates under acidic conditions (pH 4-6) including alkyl, aryl, and acyl orthophosphate monoesters and phosphorylated proteins. Has lipid phosphatase activity and inactivates lysophosphatidic acid in seminal plasma. [Isoform 2]: In addition to its tyrosine phosphatase activity, also has ecto-5'-nucleotidase activity in dorsal root ganglion (DRG) neurons. Generates adenosine from AMP. This extracellular adenosine leads to a decrease in chronic pain by activating A1R in nociceptive neurons.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Rat ACP3 antibody. ACP3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat ACP3 Antibody is added and binds to ACP3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ACP3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat ACP3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

ACP3acid phosphatase 3

View target guide

Acid phosphatase 3 (ACP3), better known as prostatic acid phosphatase (PAP or ACPP), is a human phosphatase classically associated with prostate tissue and acidic phosphomonoester hydrolysis.

Biological function: ACP3/prostatic acid phosphatase is a histidine acid phosphatase that hydrolyses phosphate monoesters under acidic conditions and can dephosphorylate phosphorylated substrates. Alternative forms of the protein include secreted and membrane-associated isoforms, supporting research into extracellular phosphatase and nucleotide-metabolism biology.
