Le kit ELISA Rat ATG3 (protéine 3 liée à l'autophagie) est un kit ELISA d'ELK Biotechnology pour la mesure de recherche de l'ATG3.
Le principe de test appliqué dans ce kit est l'immuno-dosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-revêtue d'un anticorps spécifique à l'ATG3 de rat. Des étalons ou des échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps conjugué à la biotine spécifique à l'ATG3 de rat est ajouté. Ensuite, de l'avidine conjuguée à la peroxydase de raifort (HRP) est ajoutée à chaque puits de microplaque et incubée. Après l'ajout de la solution de substrat TMB, seuls les puits contenant de l'ATG3 de rat, un anticorps conjugué à la biotine et de l'avidine conjuguée à une enzyme présenteront un changement de couleur. La réaction enzyme-substrat est terminée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré spectrophotométriquement à une longueur d'onde de 450 nm ± 10 nm. La concentration d'ATG3 de rat dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.

Biological function: ATG3 is an E2 conjugating enzyme in the autophagy machinery. It catalyses covalent conjugation of ATG8-family proteins, including LC3 and GABARAP family members, to phosphatidylethanolamine-containing membrane lipids. ATG3 cycles between ATG7-mediated loading and the ATG12-ATG5-ATG16L1 E3-like complex to promote ATG8 lipidation and formation of autophagic isolation membranes.
