ATG3 is an E2 conjugating enzyme in the autophagy machinery. It catalyses covalent conjugation of ATG8-family proteins, including LC3 and GABARAP family members, to phosphatidylethanolamine-containing membrane lipids. ATG3 cycles between ATG7-mediated loading and the ATG12-ATG5-ATG16L1 E3-like complex to promote ATG8 lipidation and formation of autophagic isolation membranes.
ATG3 is studied in LC3/GABARAP lipidation, autophagosome formation, membrane curvature sensing and autophagic flux. Assays may distinguish total ATG3 from pathway activity, so interpretation should consider nutrient or stress state, membrane context, species, application and product-level validation.
