Der Rat eIF4E (eukaryotischer Initiationsfaktor 4E) Mikromuster-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsbestimmung von eIF4E.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-eIF4E ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-eIF4E ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die Ratten-eIF4E, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-eIF4E in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
