Der Maus-MBP(Myelin Basic Protein)-Mikroproben-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsbestimmung von MBP.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für Maus-MBP spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für Maus-MBP ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Maus-MBP, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-MBP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
