Das Maus APOA4 (Apolipoprotein A4) Mikrosampel ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von APOA4.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus APOA4 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus APOA4 ist. Als Nächstes wird an Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die Maus APOA4, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus APOA4 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
