Das Rinder-TIMP2(Tissue Inhibitors Of Metalloproteinase 2) ELISA Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsbestimmung von TIMP2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Rinder-TIMP2 vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Rinder-TIMP2 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Rinder-TIMP2, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Rinder-TIMP2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
