Das Human SIRT1 (Sirtuin 1) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungs-Messung von SIRT1.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für menschliches SIRT1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der für menschliches SIRT1 spezifisch ist. Anschließend wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die menschliches SIRT1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von menschlichem SIRT1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
