Der Humane MMP7 (Matrix Metalloproteinase 7) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von MMP7 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Humanes MMP7 ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für Humanes MMP7 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Humanes MMP7, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Humanem MMP7 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
