Product description

Background

Cysteine protease required for the cytoplasm to vacuole transport (Cvt) and autophagy. Cleaves the C-terminal amino acid of ATG8 family proteins MAP1LC3, GABARAPL1, GABARAPL2 and GABARAP, to reveal a C-terminal glycine. Exposure of the glycine at the C-terminus is essential for ATG8 proteins conjugation to phosphatidylethanolamine (PE) and insertion to membranes, which is necessary for autophagy. Has also an activity of delipidating enzyme for the PE-conjugated forms (By similarity).

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Mouse ATG4B antibody. ATG4B present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse ATG4B Antibody is added and binds to ATG4B in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ATG4B antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse ATG4B. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

ATG4Bautophagy related 4B cysteine peptidase

View target guide

ATG4B (autophagy related 4B cysteine peptidase), also called cysteine protease ATG4B or autophagin-1, is an ATG4-family cysteine protease central to ATG8-family protein processing during macroautophagy.

Biological function: ATG4B is a peptidase family C54 cysteine protease that processes ATG8-family proteins, including LC3 and GABARAP family members, to expose the C-terminal glycine required for lipidation. It also removes lipid modifications from ATG8-family proteins, regulating autophagosome membrane dynamics and autophagic flux.
