L'eNOS (endothelial NOS), également connue sous le nom d'oxyde nitrique synthase 3 (NOS3) ou NOS constitutive (cNOS), est une enzyme qui, chez l'homme, est codée par le gène NOS3 situé dans la région 7q35-7q36 du chromosome 7. L'oxyde nitrique est un radical libre réactif qui agit comme un médiateur biologique dans plusieurs processus, notamment la neurotransmission et les activités antimicrobiennes et antitumorales. L'oxyde nitrique est synthétisé à partir de la L-arginine par les oxydes nitriques synthases. Les variations de ce gène sont associées à une susceptibilité aux spasmes coronariens. L'épissage alternatif et l'utilisation de promoteurs alternatifs donnent lieu à de multiples variants de transcrits.
