Le CD40 est un récepteur de surface cellulaire exprimé à la surface de toutes les cellules B matures, de la plupart des hémopathies malignes à cellules B matures et de certaines leucémies aiguës lymphoblastiques à cellules B précoces, mais il n'est pas exprimé sur les plasmocytes. Le gène CD40 est localisé sur le bras long du chromosome 20, bandes q12-q13.2. La protéine codée par ce gène est un membre de la superfamille des récepteurs du TNF. Il a été démontré que ce récepteur est essentiel dans la médiation d'une grande variété de réponses immunitaires et inflammatoires, notamment le changement de classe d'immunoglobulines dépendant des lymphocytes T, le développement des cellules B mémoires et la formation de centres germinatifs.
