La protéine codée par ce gène est un régulateur négatif du cycle cellulaire et a été le premier gène suppresseur de tumeur découvert. La protéine codée stabilise également l'hétérochromatine constitutive pour maintenir la structure globale de la chromatine. La forme active et hypophosphorylée de la protéine se lie au facteur de transcription E2F1. Des défauts dans ce gène sont une cause de rétinoblastome (RB), un cancer infantile, de cancer de la vessie et de sarcome ostéogène. [fourni par RefSeq, juillet 2008].
