La cytokératine 8 appartient à la sous-famille de type B (basique) des kératines de haut poids moléculaire et existe en combinaison avec la cytokératine 18. La cytokératine 8 se trouve principalement dans les épithéliums non squameux et est présente dans la majorité des adénocarcinomes et des carcinomes canalaires. Elle est absente dans les carcinomes épidermoïdes.
