β-Gal Senescence Staining Kit (SA-β-Gal, X-Gal)

Histochemical senescence-associated β-galactosidase (SA-β-Gal) staining kit for detecting senescence-associated enzyme activity in cells. X-Gal produces a blue visible precipitate at pH 6.0 for light-microscopy observation and counting. Qualitative staining; senescence should be interpreted alongside appropriate biological controls rather than β-Gal alone.

Specifications

• 100 nominal tests; visual/brightfield microscopy, not fluorescence.
• Kit: 100 mL staining fixative; staining solutions A and B, 1.5 mL each; staining solution C, 100 mL; X-Gal solution 5 mL.
• Adherent cells: seed to 60–80% confluence; fix 10–15 minutes; stain at 37 °C overnight without CO₂. Suspension-cell protocol also provided.
• Typical working stain per 1 mL: 10 µL A + 10 µL B + 940 µL C + 40 µL X-Gal.
• Store −20 °C protected from light, 12-month stated validity. Ice-pack transport. Product contains potentially hazardous fixative; consult safety documentation.

Technical note: avoid CO₂ incubation because pH affects SA-β-Gal assay performance. The manufacturer datasheet notes use in frozen sections may require optimisation and does not support paraffin sections.
