La SPHK1 (Sphingosine Kinase 1) est une enzyme qui, chez l'homme, est codée par le gène SPHK1. Melendez et al. (2000) ont cartographié le gène SPHK1 sur le chromosome 17q25.2 sur la base de l'identité de séquence avec des EST cartographiés dans cette région. Kohama et al. (1998) ont démontré que la Sphk1 murine recombinante peut spécifiquement phosphoryler la D-érythro-sphingosine et que la D, L-thréo-dihydrosphingosine et la N, N-diméthylsphingosine peuvent agir comme des inhibiteurs compétitifs de la Sphk1 recombinante. Pitson et al. (2000) ont découvert que la SPHK1 recombinante et la SPHK1 endogène purifiée à partir du placenta avaient des caractéristiques enzymatiques identiques, suggérant que la modification post-traductionnelle n'affecte pas les propriétés fonctionnelles.
