Mdm2 est un important régulateur négatif du suppresseur de tumeur p53. C'est le nom d'un gène ainsi que de la protéine encodée par ce gène. La protéine Mdm2 fonctionne à la fois comme une E3 ubiquitine lipase qui reconnaît le domaine de transactivation N-terminal (TAD) du suppresseur de tumeur p53 et comme un inhibiteur de l'activation transcriptionnelle de p53. Oliner et al. (1992) ont utilisé des clones MDM2 pour localiser le gène humain au chromosome 12q13-q14 par analyse d'hybrides de cellules somatiques homme-hamster.
