En utilisant des expériences de gain et de perte de fonction, il a été constaté que Xenopus Dbf4 inhibait la signalisation canonique Wnt. L'épuisement de Dbf4 endogène n'a pas perturbé les mouvements de gastrulation ou les gènes organisateurs précoces, mais a entraîné des embryons avec un cœur et des yeux morphologiquement défectueux et des marqueurs cardiaques supprimés. La fonction de Dbf4 dans le développement cardiaque semblait être indépendante de son rôle dans le cycle cellulaire. Dbf4 interagissait physiquement et fonctionnellement avec Frodo, un régulateur dépendant du contexte de la signalisation Wnt.
