Le kit ELISA de rat C3 (composant 3 du complément) est un kit ELISA de biotechnologie ELK pour la mesure de recherche du C3.
Le principe du test appliqué dans ce kit est l'immunodosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-revêtue d'un anticorps spécifique au C3 de rat. Des étalons ou des échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps biotine-conjugué spécifique au C3 de rat. Ensuite, de l'Avidine conjuguée à la Peroxydase de Raifort (HRP) est ajoutée à chaque puits de la microplaque et incubée. Après l'ajout de la solution de substrat TMB, seuls les puits qui contiennent le C3 de rat, l'anticorps biotine-conjugué et l'Avidine conjuguée à l'enzyme présenteront un changement de couleur. La réaction enzyme-substrat est terminée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré spectrophotométriquement à une longueur d'onde de 450 nm ± 10 nm. La concentration de C3 de rat dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.

C3 (complement C3) is the central complement precursor cleaved into C3a and C3b, linking complement activation to inflammatory signalling, opsonization and downstream C5 activation.
Biological function: Complement C3 is activated by C3 convertases across the classical, lectin and alternative pathways. Cleavage generates C3a, an anaphylatoxin involved in inflammatory signalling, and C3b, which covalently labels surfaces for opsonization, supports complement amplification and contributes to C5 convertase formation. Additional processing generates iC3b and other fragments with distinct immune functions.
