Product description

Background

Plays a central role in autophagy. Acts as core subunit of the PI3K complex that mediates formation of phosphatidylinositol 3-phosphate; different complex forms are believed to play a role in multiple membrane trafficking pathways: PI3KC3-C1 is involved in initiation of autophagosomes and PI3KC3-C2 in maturation of autophagosomes and endocytosis. Involved in regulation of degradative endocytic trafficking and required for the abcission step in cytokinesis, probably in the context of PI3KC3-C2. Essential for the formation of PI3KC3-C2 but not PI3KC3-C1 PI3K complex forms. Involved in endocytosis. May play a role in antiviral host defense (By similarity). Beclin-1-C 35 kDa localized to mitochondria can promote apoptosis; it induces the mitochondrial translocation of BAX and the release of proapoptotic factors.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Rat BECN1 antibody. BECN1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat BECN1 Antibody is added and binds to BECN1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BECN1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat BECN1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

BECN1beclin 1

View target guide

BECN1 (beclin 1; Beclin-1/ATG6) is a core autophagy regulator and class III PI3K-complex component linking PI3P production to autophagosome initiation and membrane trafficking.

Biological function: Beclin-1 acts as a core subunit of class III phosphatidylinositol 3-kinase complexes that generate PI3P. Distinct BECN1-containing complexes support autophagosome initiation, autophagosome maturation and endocytic trafficking, making BECN1 central to macroautophagy and membrane-remodelling pathways.
