Le kit ELISA de Rat ACSL4 (Acyl-CoA Synthetase Long Chain Family Member 4) est un kit ELISA de biotechnologie ELK pour la mesure en recherche de l'ACSL4.
Le principe du test appliqué dans ce kit est l'immunodosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-revêtue d'un anticorps spécifique de l'ACSL4 de rat. Des étalons ou des échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps conjugué à la biotine spécifique de l'ACSL4 de rat. Ensuite, de l'avidine conjuguée à de la peroxydase de raifort (HRP) est ajoutée à chaque puits de la microplaque et incubée. Après l'ajout de la solution de substrat TMB, seuls les puits contenant l'ACSL4 de rat, l'anticorps conjugué à la biotine et l'avidine conjuguée à l'enzyme présenteront un changement de couleur. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré par spectrophotométrie à une longueur d'onde de 450 nm ± 10 nm. La concentration de l'ACSL4 de rat dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.

Acyl-CoA synthetase long chain family member 4 (ACSL4), also known as FACL4 and long-chain-fatty-acid--CoA ligase 4, activates long-chain fatty acids to acyl-CoA esters. Human ACSL4 maps to NCBI Gene 2182 and reviewed UniProtKB O60488; this record links the verified target to related ABMIUM catalogue products.
Biological function: ACSL4 catalyses activation of long-chain fatty acids to fatty acyl-CoA esters for cellular lipid synthesis and fatty-acid degradation. NCBI and reviewed UniProt describe a preference for arachidonate; UniProt also identifies ACSL4 as a ferroptosis activator by generating activated polyunsaturated-fatty-acid substrates that can undergo lipid peroxidation.
