Product description

Background

Potent inhibitor of cell death. Inhibits activation of caspases. Appears to regulate cell death by blocking the voltage-dependent anion Channel (VDAC) by binding to it and preventing the release of the caspase activator, CYC1, from the mitochondrial membrane. Also acts as a regulator of G2 checkpoint and progression to cytokinesis during mitosis. Isoform Bcl-X(L) also regulates presynaptic plasticity, including neurotransmitter release and recovery, number of axonal mitochondria as well as size and number of synaptic vesicle clusters. During synaptic stimulation, increases ATP availability from mitochondria through regulation of mitochondrial membrane ATP Synthase F(1)F(0) activity and regulates endocytic vesicle retrieval in hippocampal neurons through association with DMN1L and stimulation of its GTPase activity in synaptic vesicles. May attenuate inflammation impairing NLRP1-inflammasome activation, hence CASP1 activation and IL1B release (PubMed: 17418785). Isoform Bcl-X(S) promotes apoptosis.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human BCL2L1 antibody. BCL2L1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human BCL2L1 Antibody is added and binds to BCL2L1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BCL2L1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human BCL2L1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

BCL2L1BCL2 like 1

View target guide

BCL2 like 1 (BCL2L1) encodes Bcl-2-like protein 1, whose major splice isoforms include anti-apoptotic Bcl-xL and pro-apoptotic Bcl-xS. This BCL-2-family regulator is central to mitochondrial apoptosis and cell-survival research.

Biological function: BCL2L1 produces Bcl-2-like protein 1 isoforms with opposing effects on apoptosis. Bcl-xL inhibits cell death by limiting mitochondrial cytochrome-c release and caspase activation, whereas Bcl-xS promotes apoptosis. The reviewed human protein is also implicated in cell-cycle checkpoint regulation.
