Built around the experiment

Your project. A defined technical scope.

A strong individual antibody is only the starting point. A useful antibody pair must recognise the same analyte simultaneously and produce a reliable signal in the conditions that matter to your assay.

Project context and planning

A strong individual antibody is only the starting point. A useful antibody pair must recognise the same analyte simultaneously and produce a reliable signal in the conditions that matter to your assay.

ABMIUM coordinates antigen strategy, reagent development, pairing screens and validation as one defined project. Start with new antibody development or discuss the reagents you already have. The focus is an experimentally assessed capture and detection combination, with evidence you can use to decide the next stage.

Indicative full-development timeline: approximately 5–8 months, subject to target complexity and agreed scope. Pricing is quoted after technical review.

ABMIUM reviews your specification and coordinates suitable custom-service and manufacturing routes. The proposed provider or manufacturer, available documentation and agreed scope are stated before order.

Technical scope

Start with the measurement question: which molecular form must the assay recognise, in which species and sample? Provide a gene symbol or accession, isoform, known modifications and the concentration range of interest. We review existing reagents and define acceptance criteria before selecting a development route.

A useful screening antigen should represent the form measured in your samples. Discuss full-length protein, extracellular domains, fragments or peptides, together with folding, glycosylation and other relevant modifications. Customer-supplied material can be assessed; recombinant antigen production and standards can be included when required.

The antibody strategy follows the target and intended assay. Mouse or rabbit hosts and monoclonal or polyclonal approaches can be considered during technical review. Candidate selection should preserve a range of epitope recognition rather than prioritising a single strong binder too early.

Screen combinations in both capture and detection orientations where appropriate. Assess simultaneous binding, background and interference rather than relying on individual antibody binding alone. Conjugation, immobilisation and assay buffers can change performance, so screening conditions should reflect the intended format.

Agree the experiments that will establish suitability: calibration, sensitivity, precision, working range, dilution behaviour, recovery and cross-reactivity. Matrix effects and high-dose hook behaviour may also matter. Results should identify the sample context, controls and criteria used so you can judge their relevance to your workflow.

Research applications

Sandwich ELISA

Develop a two-site measurement system with compatible capture and detection reagents. Specify the expected analyte range, available sample volume, matrix, plate format and detection chemistry. Antibody pairing and assay optimisation can be scoped together.

Lateral flow development

Assess binding reagents for a rapid assay format. Include the sample preparation, membrane or device constraints, detector label and readout method. Performance in a plate assay alone does not establish suitability for a lateral flow device.

Biomarker and specialist assays

Discuss targets with limited reagent availability or difficult biology, including low-abundance analytes, membrane-associated proteins and modified forms. Define which related proteins or isoforms the assay must distinguish, and provide representative sample information.

Project deliverables

Material, experiments and documentation are defined in the quotation. Select the deliverables that your study needs.

  • Purified capture and detection candidates at agreed quantities and formulation.
  • Recommended pair configuration and supporting screening results.
  • Calibration, specificity and agreed assay-performance reports.
  • Recombinant standard where included in the scope.
  • QC documentation, handling guidance and recommended assay conditions.
  • Optional affinity, epitope, stability and production-comparability studies.

Clear answers before you enquire

Frequently asked questions

What does a matched antibody pair do?

A capture reagent retains the analyte and a detection reagent measures the captured material. Both must bind at the same time without unacceptable competition or steric interference. Pair suitability is demonstrated in the assay, rather than inferred from two individual datasheets.

Can both antibodies be developed for the same target?

Yes, both sides of the assay can be included in the project specification. If you already have a suitable capture or detection reagent, include its clone, format, amount and available data so the missing component and pairing work can be assessed.

Do we need to supply the antigen?

No, antigen production can be discussed as a project stage. If you have material available, provide identity, purity, concentration, buffer and storage information. Its suitability depends on how closely it represents the target recognised in the intended assay.

What should we compare in a quotation?

Compare the full work package: antigen preparation, number and type of candidates, screening conditions, matrix testing, validation endpoints, final material and documentation. Clarify the milestones and what happens if agreed criteria are not met. A low entry price does not describe the total development cost.

How long does development take?

Allow approximately 5–8 months for a full development programme as an initial planning guide. An existing reagent starting point may change the route. ABMIUM confirms an indicative schedule after technical review, including dependencies such as antigen readiness and access to representative samples.

Can challenging targets be considered?

Yes. Describe the target structure, relevant modifications, species and sample context. Native epitope accessibility, antigen quality and discrimination from related molecules need particular attention. Feasibility and the proposed strategy are reviewed before work is commissioned.

Which data can be included?

Select the studies required for your decision: calibration curves, sensitivity, precision, working range, specificity and matrix performance. Affinity, epitope binning, stability and lot comparability can be discussed as additional work. Each result should be accompanied by the method and conditions used.

Can the project support scale-up?

Supply planning can be discussed during scoping, including required annual quantities, formulation, repeat production and release testing. Agree access to clones or sequences, ownership, licensing and manufacturing documentation explicitly rather than assuming these are included.

What if the first screen does not produce a usable pair?

Review the evidence against the agreed criteria before deciding on further work. Options may include different antigen material, additional candidates or revised assay conditions. Subsequent stages, costs and decision points require agreement; successful pair discovery is not guaranteed for every target.

Does this include diagnostic approval?

The service is offered for Research Use Only. A diagnostic intended use requires a separately agreed development, quality and regulatory programme. Research pairing results alone do not establish that an assay is cleared or suitable for clinical decisions.

How does antibody pair development differ from full ELISA development?

Pair development focuses on compatible capture and detection reagents. A complete ELISA programme can also include coating, detection chemistry, matrix optimisation, calibration, controls and a finished assay configuration. Agree where your project should stop so you receive the evidence and material needed for the next stage.

Can antibodies that work in Western blot be used as an ELISA pair?

Not necessarily. Western blot binding may involve a denatured target, while a sandwich assay may require simultaneous recognition of accessible epitopes on the target in solution. Assess pairing experimentally using suitable antigen and relevant assay conditions.

What affects the cost of antibody pair development?

Pricing depends on antigen readiness, antibody generation, candidate-screening depth, matrix testing and validation scope. ABMIUM provides a project-specific quotation after reviewing the requirement. Ask for a breakdown of stages, deliverables, optional studies and further-work decision points so you can compare the total scope rather than an entry price alone.
