Product description

Background

Catalyzes the initial reaction in O-linked oligosaccharide biosynthesis, the transfer of an N-acetyl-D-galactosamine residue to a serine or threonine residue on the protein receptor. Has activity toward non-glycosylated peptides such as Muc5AC, Muc1a and EA2, and no detectable activity with Muc2 and Muc7. Displays enzymatic activity toward the Gal-NAc-Muc5AC glycopeptide, but no detectable activity to mono-GalNAc-glycosylated Muc1a, Muc2, Muc7 and EA2. May play an important role in the initial step of mucin-type oligosaccharide biosynthesis in digestive organs.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human GALNT12 antibody. GALNT12 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human GALNT12 Antibody is added and binds to GALNT12 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GALNT12 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human GALNT12. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.
