Product description

Background

Involved in clearance and endocytosis of hemoglobin/haptoglobin complexes by macrophages and may thereby protect tissues from free hemoglobin-mediated oxidative damage. May play a role in the uptake and recycling of iron, via endocytosis of hemoglobin/haptoglobin and subsequent breakdown of heme. Binds hemoglobin/haptoglobin complexes in a calcium-dependent and pH-dependent manner. Induces a cascade of intracellular signals that involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production and secretion of IL6 and CSF1 (By similarity). May play a role in the process of infection of porcine monocytes/macrophages by African swine fever virus (ASFV). In case of porcine reproductive and respiratory syndrome virus (PRRSV), serves mediates virion attachment and plays a role in viral entry. After shedding, the soluble form (sCD163) may play an anti-inflammatory role.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Porcine CD163 antibody. CD163 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Porcine CD163 Antibody is added and binds to CD163 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CD163 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Porcine CD163. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

CD163CD163 molecule

View target guide

CD163 (CD163 molecule; M130) is a scavenger receptor expressed principally by monocytes and macrophages. This ABMIUM target record links human CD163 (NCBI Gene 9332; UniProt Q86VB7) to catalogue-backed research products.

Biological function: CD163 is a scavenger receptor on monocytes and macrophages that mediates clearance and endocytosis of hemoglobin-haptoglobin complexes. This pathway can limit exposure to free hemoglobin and associated oxidative damage and contributes to innate immune sensing.
