Das Ratten-CASP8(Caspase 8)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Messung von CASP8 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-CASP8 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für Ratten-CASP8 ist. Als Nächstes wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Ratten-CASP8, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-CASP8 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: Caspase-8 (CASP8; FLICE/MACH) is an initiator cysteine protease activated in death-receptor signalling complexes. It cleaves downstream caspases to promote extrinsic apoptosis and also participates in regulated cell-death and inflammatory signalling.
