Das Human BMP2 (Bone Morphogenetic Protein 2) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von BMP2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes BMP2 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann ein biotin-konjugierter Antikörper, der spezifisch für humanes BMP2 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes BMP2, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem BMP2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: Bone morphogenetic protein 2 (BMP2) is proteolytically processed to a disulfide-linked growth-factor homodimer. It initiates canonical BMP signalling through BMP type I and type II receptor complexes, including BMPR1A and BMPR2, activating SMAD-dependent transcription.
