Das Maus CD80 (T-Lymphozyten-Aktivierungsantigen CD80) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von CD80 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper gegen Maus CD80 vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein Biotin-konjugierter Antikörper, der spezifisch für Maus CD80 ist, hinzugefügt. Danach wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus CD80, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus CD80 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
