Der ziliäre neurotrophe Faktor (CNTF) ist ein potentes Polypeptidhormon, dessen Wirkungen auf das Nervensystem beschränkt zu sein scheinen, wo es das Überleben, die Neurotransmittersynthese und das Neuritenwachstum in bestimmten neuronalen Populationen fördert. Das hCNTF-Gen befindet sich auf Chromosom 11, wie mithilfe von menschlich-hamsterischen Somazellhybriden festgestellt wurde. Das CNTF-Protein ist in der Evolution hoch konserviert. Die Aminosäure-(aa)-Sequenzen von Ratten- und Kaninchen-CNTF, die aus cDNAs übersetzt wurden, zeigen ca. 85 % Homologie mit der abgeleiteten aa-Sequenz, die hCNTF kodiert. CNTF induziert Gewichtsverlust und verbessert die Glukosetoleranz bei Menschen und Nagetieren. Es wird angenommen, dass CNTF zentral wirkt, indem es die hypothalamische Neurogenese induziert, um die Nahrungsaufnahme zu modulieren, und peripher, indem es die hepatische Genexpression auf ähnliche Weise wie Leptin verändert.
