CD44 ist ein integrales Zellmembran-Glykoprotein mit einer postulierten Rolle bei der Matrixadhäsion, Lymphozytenaktivierung und Lymphknoten-Homing. Es enthält 19 Exons, die 50 kb genomische DNA umfassen. Beim Menschen wird das CD44-Antigen durch das CD44-Gen auf Chromosom 11 kodiert. Das von diesem Gen kodierte Protein ist ein Zelloberflächen-Glykoprotein, das an Zell-Zell-Interaktionen, Zelladhäsion und Migration beteiligt ist. Es ist ein Rezeptor für Hyaluronsäure (HA) und kann auch mit anderen Liganden wie Osteopontin, Kollagenen und Matrix-Metalloproteinasen (MMPs) interagieren. Transkripte für dieses Gen durchlaufen ein komplexes alternatives Spleißen, das zu vielen funktionell unterschiedlichen Isoformen führt, wobei jedoch die vollständige Länge einiger dieser Varianten noch nicht bestimmt wurde. Alternatives Spleißen ist die Grundlage für die strukturelle und funktionelle Vielfalt dieses Proteins und kann mit der Tumormetastasierung zusammenhängen.
