R-spondin-1 ist ein sezerniertes Protein, das beim Menschen durch das Rspo1-Gen kodiert wird. Es ist auf Chromosom 1p34 kartiert. Dieses Gen kodiert ein sezerniertes Aktivatorprotein mit zwei cysteinreichen, furinähnlichen Domänen und einer Thrombospondin-Typ-1-Domäne. Das kodierte Protein ist ein Ligand für Leucin-reiche Wiederholungs-haltige G-Protein-gekoppelte Rezeptoren (LGR-Proteine) und reguliert positiv den Wnt-Signalweg. Bei Mäusen induziert das Protein das schnelle Einsetzen der Kryptenzellproliferation und erhöht die intestinale Epithelheilung, wodurch ein schützender Effekt gegen chemotherapieinduzierte Nebenwirkungen erzielt wird. Alternatives Spleißen führt zu mehreren Transkriptvarianten.
