CD28 (Cluster of Differentiation 28) ist eines der Proteine, die auf T-Zellen exprimiert werden und kostimulatorische Signale liefern, die für die T-Zell-Aktivierung und das Überleben erforderlich sind. Die T-Zell-Stimulation durch CD28 zusätzlich zum T-Zell-Rezeptor (TCR) kann ein potentes Signal für die Produktion verschiedener Interleukine (insbesondere IL-6) liefern. Das CD28-Gen ist auf 2q33-q34 kartiert. CD28 und CTLA-4 bilden zusammen mit ihren Liganden B7-1 und B7-2 einen der dominanten kostimulatorischen Signalwege, die T- und B-Zell-Antworten regulieren. CD28 und CTLA-4 sind strukturell homologe Moleküle, die Mitglieder der Immunglobulin (Ig)-Gen-Superfamilie sind.
