Der ELISA Kit für menschliches ADAMTS1 (A Disintegrin And Metalloproteinase With Thrombospondin 1) ist ein ELK Biotechnology ELISA Kit zur Messung von ADAMTS1 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes ADAMTS1 ist. Standards oder Proben werden dann zusammen mit einem biotin-konjugierten Antikörper, der spezifisch für humanes ADAMTS1 ist, in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes ADAMTS1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem ADAMTS1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
