Dieses Gen gehört zur Familie der Homeobox-Gene. Die Homeobox-Gene kodieren eine hochkonservierte Familie von Transkriptionsfaktoren, die eine wichtige Rolle bei der Morphogenese in allen vielzelligen Organismen spielen. Säugetiere besitzen vier ähnliche Homeobox-Gencluster, HOXA, HOXB, HOXC und HOXD, die sich auf verschiedenen Chromosomen befinden und aus 9 bis 11 Genen bestehen, die hintereinander angeordnet sind. Dieses Gen ist eines von mehreren Homeobox-HOXD-Genen, die sich in den Chromosomenregionen 2q31-2q37 befinden. Deletionen, die den gesamten HOXD-Gencluster oder das 5'-Ende dieses Clusters entfernt haben, wurden mit schweren Gliedmaßen- und Genitalanomalien in Verbindung gebracht.
