Die präzise Transkriptionsinitiierung bei TATA-haltigen Genen der Klasse II beinhaltet den geordneten Zusammenbau der RNA-Polymerase II (POLR2A; MIM 180660) und der allgemeinen Initiationsfaktoren TFIIA, TFIIB (MIM 189963), TFIID (MIM 313650), TFIIE (MIM 189962), TFIIF (MIM 189968), TFIIG/TFIIJ und TFIIH (MIM 189972). Der erste Schritt beinhaltet die Erkennung des TATA-Elements durch die TATA-bindende Untereinheit (TBP; MIM 600075) und kann durch TFIIA reguliert werden, einem Faktor, der sowohl mit TBP als auch mit einem TBP-assoziierten Faktor (TAF; MIM 600475) in TFIID interagiert. TFIIA hat 2 Untereinheiten (43 und 12 kD) in Hefe und 3 Untereinheiten in höheren Eukaryoten.
