Dieses Gen kodiert ein klassisches Cadherin der Cadherin-Superfamilie. Alternatives Spleißen führt zu mehreren Transkriptvarianten, von denen mindestens eine ein Präproprotein kodiert, das proteolytisch verarbeitet wird, um das reife Glykoprotein zu erzeugen. Dieses kalziumabhängige Zell-Zell-Adhäsionsprotein besteht aus fünf extrazellulären Cadherin-Wiederholungen, einer Transmembranregion und einem hochkonservierten zytoplasmatischen Schwanz. Dieses Gen befindet sich in einem Gencluster in einer Region auf dem langen Arm von Chromosom 16, die an Loss-of-Heterozygozität-Ereignissen bei Brust- und Prostatakrebs beteiligt ist. Darüber hinaus wird eine aberrante Expression dieses Proteins bei zervikalen Adenokarzinomen beobachtet. Mutationen in diesem Gen sind mit Hypotrichose mit juveniler Makuladystrophie und ektodermaler Dysplasie, Ektrodaktylie und Makuladystrophie-Syndrom (EEMS) assoziiert.
