Hintergrund

Ceramide werden während der epidermalen Differenzierung synthetisiert und reichern sich in den Zwischenräumen des Stratum corneum an, wo sie kritische Komponenten der epidermalen Permeabilitätsbarriere darstellen. Überschüssiges zelluläres Ceramid kann antimitogene Signale auslösen und Apoptose induzieren, und die Ceramidmetaboliten Sphingosin und Sphingosin-1-phosphat (S1P) sind wichtige bioregulatorische Moleküle. Die Ceramidhydrolyse in den kernhaltigen Zellschichten reguliert die Keratinozytenproliferation und Apoptose als Reaktion auf externen Stress. Die Ceramidhydrolyse findet auch im Stratum corneum statt, wodurch freie Sphingoidbase freigesetzt wird, die als endogenes antimikrobielles Mittel fungiert. ACER1 wird in der Epidermis stark exprimiert und katalysiert die Hydrolyse von sehr langkettigen Ceramiden zur Generierung von Sphingosin (Houben et al., 2006 [PubMed 16477081]; Sun et al., 2008 [PubMed 17713573]).[bereitgestellt von OMIM, Juli 2010],
