Galanin ist ein wichtiger Neuromodulator, der im Gehirn, im Magen-Darm-System und in der Hypothalamus-Hypophysen-Achse vorkommt. Es ist ein 30 Aminosäuren langes, nicht-C-terminal aminiertes Peptid, das die Wachstumshormonsekretion stark stimuliert, die kardiale vagale Verlangsamung der Herzfrequenz hemmt, die Sinusarrhythmie aufhebt und die postprandiale gastrointestinale Motilität hemmt. Die Wirkungen von Galanin werden durch die Interaktion mit spezifischen Membranrezeptoren vermittelt, die Mitglieder der 7-Transmembran-Familie von G-Protein-gekoppelten Rezeptoren sind. GALR2 interagiert mit den N-terminalen Resten des Galaninpeptids. Der primäre Signalmechanismus für GALR2 erfolgt über den Phospholipase C/Proteinkinase C-Weg (über Gq), im Gegensatz zu GALR1, das sein intrazelluläres Signal durch Hemmung der Adenylylzyklase über Gi kommuniziert.
