Rekombinantes humanes Trophoblast-Glykoprotein wird von unserem Säugetier-Expressionssystem produziert, und das Zielgen, das Ser32-Ser355 kodiert, wird mit einem 6His-Tag am C-Terminus exprimiert.
Trophoblast-Glykoprotein, auch bekannt als TPBG und 5T4, gehört zur LRR-Proteinfamilie. Der C-Terminus der LRR N-terminalen Kappe (LRRNT) und LRR 1 sind essenziell für die Hemmung des Wnt-Signalwegs. TPBG könnte als Inhibitor der Wnt/Beta-Catenin-Signalübertragung fungieren, indem es indirekt mit LRP6 interagiert und die Wnt3a-abhängige LRP6-Internalisierung blockiert. Es wird von allen Trophoblastentypen bereits ab der 9. Entwicklungswoche exprimiert. In erwachsenen Geweben ist die Expression auf einige wenige Epithelzelltypen beschränkt, wird aber bei einer Vielzahl von Karzinomen gefunden.
