Rekombinantes humanes Vitamin-D-bindendes Protein wird von unserem Säugetier-Expressionssystem produziert und das Zielgen, das Leu17-Leu474 kodiert, wird mit einem 6His-Tag am C-Terminus exprimiert.
Vitamin-D-bindendes Protein (DBP) ist ein Mitglied der ALB/AFP/VDB-Familie. DBP ist ein sezerniertes Protein und enthält drei Albumin-Domänen. Die Primärstruktur enthält 28 Cysteinreste, die mehrere Disulfidbrücken bilden. DBP fungiert als multifunktionales Protein, das in Plasma, Aszitesflüssigkeit, Liquor cerebrospinalis und Urin sowie auf der Oberfläche vieler Zelltypen gefunden wird. DBP bindet Vitamin D und seine Plasmametaboliten und transportiert sie zu den Zielgeweben. DBP assoziiert mit membrangebundenem Immunglobulin auf der Oberfläche von B-Lymphozyten und mit dem IgG-Fc-Rezeptor auf den Membranen von T-Lymphozyten.
