Autophagie wird im Allgemeinen durch Nährstoffmangelbedingungen aktiviert, wurde aber auch mit einer Reihe physiologischer Prozesse in Verbindung gebracht, darunter Entwicklung, Differenzierung, Neurodegeneration, Infektion und Krebs. Die Bildung des Autophagosoms beinhaltet ein Ubiquitin-ähnliches Konjugationssystem, bei dem Atg12 kovalent an Atg5 gebunden und zu Autophagosom-Vesikeln geleitet wird. Diese Konjugationsreaktion wird durch das Ubiquitin-E1-ähnliche Enzym Atg7 und das E2-ähnliche Enzym Atg10 vermittelt.
